Rapamycin Improves Brain Blood Flow in APOE4 Carriers

A four‑week trial gave a low daily dose of rapamycin to adults aged 45‑65. Participants with the APOE4 variant showed measurable increases in cerebral blood flow, while non‑carriers did not. Women with APOE4 had the largest benefit, suggesting sex‑specific effects.

These data point to rapamycin as a candidate for early intervention in people genetically predisposed to Alzheimer’s, but the study did not test long‑term cognitive outcomes. The effect was limited to a small sample and short duration, and whether improved circulation translates to reduced disease risk remains unknown. Clinicians and patients should view these results as preliminary.
